When responding to job leads, your cover note (generally your email) is extremely important. It should point the reviewer to a couple of specific spots on your resume that are key to the job.As an example, a posting for Corporate Controller may specify previous employment at a public company and experience with SEC reporting are required. The recruiter may do the typical 15 second scan of you resume (he/she has 100 resumes to review) and not spot the appropriate experience. But a brief cover note that points out that "while Controller at ABC Company from 2001 to 2004, I was responsible for all SEC reporting" would direct the recruiter to the right spot on the resume. Of course the resume should also make this clear. But if the recruiter misses it, your resume will likely be discarded, even though you have the right experience.
